Prepare samples and groups
Open the project’s Samples tab. Add samples from an existing dataset or start an import, then create groups such as Control and Treatment.
Create groups
Section titled “Create groups”Create at least two groups for a comparison. Move biological replicates into the correct group and keep technical replicates together according to your experimental design. Rename groups with meaningful condition names before continuing.
Choose a baseline deliberately
Section titled “Choose a baseline deliberately”The baseline group is the reference. A comparison of Treatment against Control reports positive changes as higher in Treatment and negative changes as lower in Treatment. Reversing the baseline reverses that interpretation.
Before continuing
Section titled “Before continuing”Confirm every sample is assigned once, group sizes are sensible, paired reads are present where expected, and the organism matches the project. Continue to run setup only after the grouping board reflects the experiment you intend to defend.
